Tumor-derived nucleoli preparation (TN) can be easily prepared from human hepatomas. Nucleoli from various tumors have a common structural feature. Therefore, we examined whether TN could be used as a tissue source for the production of an authentic protein. Using an in vitro system, which contains both microsomes and TN as post-ribosomal supernatant, we demonstrated that TN contains more and larger polyribosomes than microsomes, and can catalyze the synthesis of authentic alpha-fetoprotein (AFP) in vitro. Furthermore, it was shown that the translation rate from the alpha-fetoprotein mRNA in the TN system is approximately 3 times higher than that in a microsomal system.
